Termination of pregnancy and induction of premature luteolysis by the antiprogestagen, mifepristone, in dogs.

چکیده
Five pregnant beagle bitches were treated with 2.5 mg mifepristone/kg body weight, twice a day, for 4.5 days starting at Day 32 of gestation. Results of fetal ultrasonography and assay of serum progesterone concentrations every 2-4 days were compared to those in 5 control bitches. Mifepristone resulted in a premature (P less than 0.01) termination of pregnancy (36 +/- 1 vs 65 +/- 1 days), without side effects. The antiprogestagen also caused progesterone to decline to less than 1 ng/ml by Day 40-45 after the preovulatory LH peak (vs 64-67 days in controls) and reduced (P less than 0.05) mean concentrations on Days 34-50 (2.2 +/- 0.5 vs 6.3 +/- 0.3 ng/ml). The results suggest that antiprogestagen therapy is a safe means to terminate unwanted pregnancy in dogs, and that luteal function in pregnant bitches is dependent on luteotrophic support that is blocked by antiprogestagen treatment, directly or indirectly, due to termination of pregnancy.

Induced Termination of Pregnancy in Domestic Farm Animals
Induced abortion may be desired during a normal or abnormal pregnancy. The methods of induced abortion differ between species mainly because of different sources for progesterone production during pregnancy between species and within a species according to different stages of gestation. In cows and buffaloes, pregnancy can be effectively terminated from 5 to 150 days by administration of luteol...
متن کاملInfluence of Mifepristone in Induction Time for Terminations in the Second and Third Trimester.
Termination of pregnancy after the first trimester is generally carried out by medical induction. Question: The aim of this study is to investigate the effect of mifepristone before administration of the prostaglandin derivative on induction time. Material and Methods: We analysed 333 medically indicated terminations after the first trimester under the terms of § 218 a Para. 2 of the German Cri...
متن کاملO-31: Mifepristone Acts as Progesterone Antagonistof Non-Genomic Responses but InhibitsPhytohemagglutinin Induced Proliferationin Human T Cells
Background: Progesterone is an endogenous immunomodulator that suppresses T cell activation during pregnancy. The stimulation of membrane progesterone receptors (mPRs) would seem to be the cause of rapid non-genomic responses in human peripheral T cells, such as an elevation of intracellular calcium ([Ca2+] i) and decreased intracellular pH (pHi). Mifepristoneimmune cells compared with progeste...

Background & Aim: Premature rupture of membrane is one of the most common problems in obstetrics, and successful labor induction, especially in cases with low bishop score(BS), is an important factor in decreasing maternal and fetal complications. The object of this study was to assess the efficacy and safety of vaginal misoprostol on duration of labor in preterm premature rupture of membra...
متن کاملA pilot study on the use of letrozole with either misoprostol or mifepristone for termination of pregnancy up to 63 days.
BACKGROUND Letrozole is a third-generation selective aromatase inhibitor. Animal data suggested that it might be useful in medical abortion. We performed two pilot studies to assess the feasibility of using letrozole in combination with either mifepristone or misoprostol for termination of pregnancy up to 63 days. STUDY DESIGN We recruited 40 subjects who requested legal termination of pregna...
